Bhupendra Patel was a member of Memnagar Municipality from 1995 to 1996, 1999 to 2000 and 2004 to 2006. In 2008–10 he served as the Chairman of the Ahmedabad School Board. In 2012, Bhupendra Patel introduced the idea of reducing fees for builders to incentivize the building regulation scheme and received praise from then Chief Minister Narendra Modi for this initiative. He served as the councilor of Thaltej ward from 2010 to 2015. During the same period, Bhupendra Patel also served as the Chairman of the Standing Committee of the Ahmedabad Municipal Corporation. From 2015 to 2017, he served as the Chairman of Ahmedabad Urban Development Authority (AUDA). During his tenure at AUDA, he spearheaded infrastructure projects including flyovers and roads in Sanand and Bopal. In 2017, he was elected MLA in the Gujarat Legislative Assembly election from Ghatlodiya constituency. Before him there was Anandiben Patel. In these elections, Bhupendra Patel defeated his rival, Shashikant Patel of the Indian National Congress, by a margin of 1,17,000 votes.

On 12 September 2021, he was elected as the Chief Minister of Gujarat. Former Chief Minister of Gujarat Vijay Rupani resigned from the post of Chief Minister of Gujarat on 11 September 2021. In an interview with BBC Media, journalist and political analyst Faisal Bakili said,
Perhaps due to speculations that there is an anti-incumbency wave against the BJP government in Gujarat, the party has chosen a new face as the Chief Minister.
On 13 September 2021, he was sworn in as the seventeenth Chief Minister of Gujarat.
On 8 December 2022, he won his second consecutive victory in the Gujarat Assembly elections from the urban constituency of Ghatlodiya by a margin of 1.92 lakh. On 12 December 2022, he took oath as the Chief Minister of Gujarat for the second time.
