In 1990, Premalatha Vijayakanth married actor-turned-politician Vijayakanth in Madurai. Dravida Munnetra Kazhagam leader M. Karunanidhi attended their wedding. Once while talking to the media, he had told that his father was influenced by Vijaykant and that is how his marriage was fixed. He said,
When he came to meet me, my father was immediately impressed by his attitude. He thought that if he wanted to give his daughter, he should give it to this man. He then came across not as a hero but as an ordinary man.”
The couple has two sons named Shanmuga Pandian and Vijay Prabhakar. Shanmuga Pandian is an actor.

When DMDK was founded in 2006, Premalatha Vijayakanth started supporting her husband’s party and gained popularity in Tamil Nadu through her platform speeches. On 19 October 2018, he was appointed as the DMDK party treasurer. He addressed issues like power cuts, price rise and law and order concerns and identified the problems faced by people in different areas.

In the 2021 Tamil Nadu Assembly election, Premalatha Vijayakanth contested and won from the Vriddhachalam constituency. Later, at the 18th Executive and General Council meeting of the DMDK in December 2023, her husband Vijayakanth announced that she would serve as the general secretary of the party as she was not well.
