In April 2003, he won kickboxing at the Fast a Fearless event. Later, he was brought to the UFC and made his debut against Tim Means on April 20, 2013 at UFC on Fox 7, where he won the fight by unanimous decision. He faced Rustam Khabilov in November 6, 2016 but lost the back and forth fight. However, they earned Masvidal his first Fight of the Night. In July 26, 2014, faced Daron Cruickshank and won the fight. After that, he faces James Krause and wins the fight.
In July 12, 2015, faced Cezar Ferreira in a welterweight bout at The Ultimate Fighter 21 Finale, where he won the fight via first-round knockout, also earning a Performance of the Night bonus. 2016-present

He retired Cerrone and nearly finished the fight in the final seconds of the first round, eventually winning the fight via TKO in the second round after scoring another drop. The win even gave Masvidal his second Performance of the Night bonus.
On May 13, 2017, he faced Demian Maia at UFC 211 but lost the fight via split decision. Likewise, he faced Ben Askren on July 6, 2019 at UFC 239. He even won the fight via a flying knee 5 seconds into the first round. This victory earned him the 2019 Performance of the Night and Knockout of the Year award from various MMA media outlets. Consequently, he signed a new eight-fight contract with the UFC.
